L04: Chronic kidney disease - a clinical model of premature aging

Aging is not only associated with increased wisdom and experience, but also with a decline in physical functions including vascular calcification, heart failure, osteoporosis, muscle loss, frailty, depression and cognitive disorders. CKD with accumulation of uremic toxins and increased modification of proteins leads to premature occurence of aging. Prof. Stenvinkel, chairman of the Life/2021 conference has dedicated his work to understand the molecular and physiological mechanisms behind this phenomenon and thereby create a foundation for treatments and diagnostics.

Presently available information suggests that Online hemodiafiltration (HDF) is associated with an improved clinical outcome when compared to standard hemodialysis. However, definite proof is lacking and information on the effects on patient reported outcome measures is very limited. Prof. Blankestijn, principal investigator of CONVINCE will share insights and current status of this global clinical trial, which is supported by the Horizon 2020 program of the European Commission.

Despite the tremendous efficacy of several new drug classes such as SGLT2-inhibitors, endothelin receptor antagonists, and mineralocorticoid receptor antagonists on a population level, not every patient benefits to the same extent. Understanding the underlying mechanisms for the variation in drug response will pave the way for personalized therapy approaches. Prof. Lambers-Heerspink is maintainer of personalized medicine by optimizing and finding new therapeutic approaches to halt the progression of renal and cardiovascular diseases in patients with diabetes.
